Summary
These tau knockout rats were created through transcription activator-like effector nuclease (TALEN)-mediated targeting of exon 2 of the rat Mapt gene (Ayers, Lopez et al., 2024).
Tau protein was undetectable in the brains of Cy23 rats by western blotting or immunohistochemistry using the T49 antibody selective for rodent tau.
No gross phenotypes were observed in rats up to 24 months of age.
Cy23 rats have been intercrossed with Tg12099 rats that express a human MAPT transgene with a mutation linked to frontotemporal dementia in order to study the effects of human tau in the absence of rodent tau.
Absence of tau in Mapt knockout (Cy23) rats. Left, western blot of brain homogenates from wild-type (+/+), heterozygous (+/0) and Mapt knockout (-/-) rats, probed using the T49 antibody selective for rodent tau. Right, sections through the brains of wild-type (Mapt +/+) and Mapt knockout (Mapt 0/0) rats, immunostained with T49. From Ayers, Lopez et al., 2024; licensed under Creative Commons BY 4.0.
