This very interesting paper continues the work of the Schon laboratory investigating the role that the mitochondrial-associated membranes (MAM) of the endoplasmic reticulum may play in Alzheimer's disease. Schon and co-workers have previously shown that the presenilin proteins and γ-secretase activity are concentrated in MAM and they observed increases in MAM in fibroblasts from both sporadic and familial AD individuals. In this paper they show that conditioned media from mouse astrocytes expressing human ApoE4 increases MAM activity in fibroblasts relative to that seen for treatment with conditioned media from astrocytes expressing human ApoE3. These observations increase the support for the idea that changes in MAM activity may play a central role in AD pathogenesis.
