Role of Intracellular Aß42 in AD
This makes it attractive to postulate that fibril formation begins intracellularly
- There clearly is Aß in cells, particularly in neurons.
- Aß42 appears to comprise a higher proportion of intracellular than extracellular Aß.
- Although rigorous quantitation of intracellular Aß is challenging, the emerging data suggest that the intracellular concentration of Aß1-42 is far higher than the extracellular concentration, and likely to be in the range required for fibril formation.
- The reduced intraluminal pH found in cellular organelles favors fibril formation.
