Transgene: A 2.3 kb cDNA fragment containing the entire coding region of the (h)SIRT1
was inserted into a unique HindIII-Not I restriction site located within the second
exon of the rat neuron-specific enolase (rNSE) gene.Promoter: rat neuron specific
enolase (NSE) promoter, including 2600 bp segment of promoter and 5' flanking region,
as well as exon 1, intron 1, and 6 bp of exon 2 of rat NSE gene.

Mouse strain: C57Bl/6J x SJL/J (B6SJL) F1
Neuropathological Analysis:
~ 2 fold elevation of hSIRT1 expression was found in cortex of ~ 3 months-old (h)SIRT1,
relative to gender-matched wild type (WT) littermate controls. A significant diminution
of ROCK1 protein content (p<0.05) in the same brain tissue was also found, relative
to gender-matched wild type (WT) littermate controls. We also detected a commensurate
elevation of a-secretase activity (P<0.05) in the cortex of (h) SIRT1 trangenics,
relative to gender-matched wild type (WT) littermate controls.
