Transgene: The coding sequence for human mutant [A30P] αSYN was cloned into
a cassette containing 4.5-kb 5' flanking DNA from the rat TH gene using NheI
and SpeI. The linearized construct was injected into fertilized DBA X C57BL/6
eggs.
Promoter: Rat TH promotor
Mouse Strain: Backcrossed into the C57BL/6 strain for 4 generations (2001)
Neuropathological Analysis:
Tg mice showed somal and neuritic accumulation of transgenic [A30P] αSYN in
TH-positive neurones in the substantia nigra. However, there was no difference in
the number of TH-positive neurones in the substantia nigra and the concentrations
of catecholamines in the striatum between these transgenic mice and non-transgenic
littermates.
