PubMed Abstract
This is an important article because it validates the relationship between CAA and hemorrhage, which was one of the earliest APP related mutations associated with dementia.
REAGENTS/MATERIAL: Generation of B6,D2-TgN (Thy1-APPSwe)23 Trasgenic mice as described in Sturchler-Pierrat et al).
APP23 mice overexpress APP751 with the Swedish double mutation under the control of a neuron-specific Thy-1 promoter element, backcrossed with C57BL/6J mice. Cresyl violet, hematoxylin and eosin (H&E), and Congo red staining were done.
Avidin-biotin-peroxidase complex method (Vector Laboratories) with diaminobenzidine as chormogen.
Polyclonal antibody AS42/14 specifically to Ab42
Polyclonal antibody FCA3340 and FCA3542 specifically to Ab40 and Ab42, respectively, gift from F. Checlair.
Mouse monoclonal antody to a-smooth muscle actin, clone 1A4; (Sigma)
Mouse monoclonal antibody to b-dystroglycan (Novocastra)
Polyclonal antibody to glial fibrillary acidic protein (GFAP) (Dako)
Polyclonal antibody to cystatin C (Accurate Chemicals)
Polyclonal antibody to mouse serum amyloid P component (SAP) (Calbiochem)
Polyclonal antibody to AB (NT12)
Secondary antibodies, Alexa 568 goat anti-rabbit IgG, Alexa 488 goat anti-mouse IgG (Molecular Probes)
FUTURE DIRECTION: Further evaluation of CAA in AD dementia needed. Vaccination in APP23 mice might be a good study tool on CAA since PDAPP transgenic mice do not develop significant CAA.
