REAGENTS/MATERIAL: Antibodies used in this study:
BACE1 was detected using rabbit anti-BACE1 [residues 46-163]
and 7523 [residues 46-60]
(provided by Dr. Christian Haass, Ludwig-Maximilians-University, Munich), or monoclonal
anti-BACE1-Cat1 [residues 46-460]
and anti-FLAG M2 (Sigma).
Rabbit polyclonal antiserum CTM1 was raised against a synthetic peptide corresponding to the C-terminal 15 amino acids of APP followed by the c-Myc
epitope (MEQKLISEEDLN). Rabbit polyclonal anti-PS1NT (residues 1-65) has been
described.
Both mAb anti-Aβ 26D6
and anti-Aβ (B436)
(epitope within residues 1-12 of Aβ) react with the amino-terminal region of Aβ and also recognize sAPPα; mAb B113 is selective for Aβ40.
The following antibodies were purchased from commercial sources:
monoclonal mouse anti-GM130 (35) (BD Biosciences);
monoclonal mouse anti-syntaxin 6 (30) (BD Biosciences);
monoclonal mouse anti-flotillin-2 (29) (BD Biosciences);
monoclonal mouse anti-γ-adaptin (88) (BD Biosciences);
monoclonal mouse anti-EEA1 (14) (BD Biosciences);
monoclonal mouse anti-HA (16B12) (Covance Research Products);
monoclonal mouse anti- Aβ (4G8) (Covance) and
monoclonal mouse anti-APP, secretory (OM84) (Chemicon Millipore).
