REAGENTS/MATERIAL:
Proteins from the postnuclear supernatants were used for co-immunoisolation using PS1_NT antibody as described previously (paper). Antibodies against γ-secretase subunits, APP, and organelle markers have been described (paper). A goat anti-CD147 (Santa Cruz Biotechnology) and a mouse monoclonal anti-human CD147 (1G6.2) (Chemicon) were used to detect CD147. Immunofluorescence staining as well as isolation of DRM from Lubrol WX lysates of cultured cells by discontinuous flotation density gradients were performed as described (paper, paper). Immunohistochemical staining was performed on 30 μm sagittal sections of 2 month-old C57BL/6J mouse brain with rabbit anti-nicastrin (SP718, raised against synthetic peptides corresponding to residues 689-709 ) or goat anti-CD147 antibodies as described (paper)
