REAGENTS/MATERIAL:
Western blots were probed with primary antibody against BACE1 (PA1-757; 1:1000 dilution; Affinity BioReagents, Golden, CO), APP (22C11; 1:5000 dilution; Chemicon, Temecula, CA), APPs(sw) neoepitope (1:5000 dilution) (Seubert et al., 1993; Vassar et al., 1999; Cole et al., 2005), or b-actin (1:15000 dilution; Sigma). Ab40 levels in hemibrain homogenates were determined using an Ab40-specific sandwich ELISA (BioSource International, Camarillo, CA) according to the recommendations of the manufacturer. For IHC brain sections were incubated with rabbit anti-glial fibrillary acidic protein (GFAP) primary antibody (1:10,000; Sigma).
