Western blots were probed with rabbit monoclonal anti-cleaved caspase-3 antibody (5A1, Cell Signaling) dilutions at 1:1000 for gastrocnemius and testes, 1:300 for thymus, and 1:1000 for duodenum.

Produced D257A mtDNA mutant mice that display premature aging phenotypes. The mice have a reduced life span and thymic involution, testicular atrophy associated with the depletion of spermatogonia, loss of bone mass, loss of intestinal crypts, progressive decrease in circulating red blood cells, and weight loss.
