X11b transgenic mice were developed using mouse prion protein promoter and a carboxyl-terminal Myc-tagged full-length mouse X11b cDNA. APPswe Tg2576 mice were used for crossing with X11b mice.
Mouse brains were immunostained for X11b by using a rabbit X11 polyclonal antibody or mouse monoclonal antibody 9B11 to the Myc tag on the transgene; Ab deposits were detected using antibody 1E8.
Ab species were assayed using commercial ELISA kits and following the manufacturers' instructions. Ab(1-40) was assayed using humanAb(1-40)-(N) ELISA (IBL), and Ab(1-42) was detected using Innotest b-amyloid(1-42) ELISA (Innogenetics).
