MnSOD heterozygous knockout mice were obtained from Dr Charles Epstein (University of California, San Francisco). Tg19959 mice, which overexpress a doubly mutated human b-amyloid precursor protein (APP)(KM670/671NL and V717F) were produced, backcrossed to C57BL6 then mated with MnSOD mice.
The following antibodies were used for immunohistochemistry: anti-Ab40 and anti-Ab42 antibodies (1:1000, Chemicon); 6E10 and 4G8 (1:1000, Signet Laboratories), and anti-MnSOD antibody (1:1000, Stressgen Biotechnologies). Western blots were probed with 6E10 and protein loading was confirmed with anti-atubulin antibody (1:10,000, Sigma).
