REAGENTS/MATERIAL:

Tg2576 mice were used in this study.

The following antibodies were used: BAN-50 (anti-Ab1–16), 4G8 (anti-Ab17–24, Senetek); 3160 (anti-Ab1–40) for immunoprecipitation (Kawarabayashi et al., 2001); O443 (C-terminal 20 amino acid residues of APP, a gift from K. Sambamurti); Z31preA [antibody to 577–596 of APP695 (Kawarabayashi et al., 1991)]; anti-flotillin antibody (Transduction Laboratories), anti-calnexin antibody (StressGen Biotechnologies); anti-mouse ApoE antibody (a gift from N. Yamada), anti-human ApoE antibody (Chemicon); PHF-1 to phosphorylated tau at pSer396/pSer404 sites, and WKS46 as a phosphorylation-independent antibody against tau 359–37 (gift from S. H. Yen), antibody to N terminus of mouse PS1 (N1) and antibody to loop of mouse PS-1 (L1) (gifts from T. Koothan); anti-BACE-1 antibody (rb206; Amersham Biosciences), and anti-neprilysin antibody (CD10; Novocastra Laboratories).

ELISA assay employed BNT-77/BA-27 for Ab40 and BNT-77/BC-05 for Ab42 (Suzuki et al., 1994; Shinkai et al., 1997).
