REAGENTS/MATERIAL:

Western blots probed with polyclonal antibody anti-APP369 (kindly provided by Sam Gandy, Thomas Jefferson University, Philadelphia, PA, USA), which is specific to the C-terminus of APP.

Mouse brain homogenates were subjected to an ELISA with capture monoclonal antibody BNT77 (specific to Ab 17–28) and monoclonal detection antibodies BA27 for Ab40 and BC05 for Ab42. For determination of levels of Ab40 at various ages of mice, a commercially available ELISA was utilized (Biosource International, Camarillo, CA, USA).

Ab peptides were immunoprecipitated with monoclonal antibodies 4G8 or 6E10 (Signet Laboratories, Dedham, MA, USA) using protein A/G-plus agarose beads.

for immunohistochemistry monoclonal antibody 6E10 to Ab 1–17 (Signet Laboratories, Dedham, MA, USA) and the amyloid-specific dye Thioflavine-S were utilized to visualize Ab deposits in brain sections.
