  Primary News: Second Study to Confirm IDE-Amyloid Connection in Mice

The newly published paper by Miller et al., well summarized by Tom Fagan above, both confirms and complements our recent study, in which we also utilized IDE gene-disrupted mice to demonstrate that IDE regulates the levels of cerebral Aβ and the AβPP intracellular domain (AICD, CTF in vivo: see Farris et al., 2003). A major strength of this latest work is that by breeding heterozygous IDE knockout mice (IDE+/-) to each other, these authors generated single litters containing all three possible IDE genotypes (IDE+/+, IDE+/-, and IDE-/-). The advantages of this breeding strategy are that it enables one to look for a "gene-dosage" effect on the phenotype of interest while providing littermates of different genotypes for comparison. The disadvantage of this method is that relatively few mice of a given genotype can be generated within one litter. REAGENTS/MATERIAL:

Studied insulysin-deficient (IDE) mice.

Prepared rIDE4020 antibody by immunization with nickel agarose-purified recombinant rat insulysin with an N-terminal six-histidine tag, which was expressed by a baculovirus system in Sf9 insect cells. The hIDE7-24 antibody was elicited by immunization with a synthetic peptide that corresponds to amino acids 7-24 (relative to the second ATG) of human insulysin that was coupled to keyhole limpet hemocyanin by a C-terminal cysteine residue. These antibodies were used for immunodetection.

Sandwich ELISA for Ab40 and Ab42 were performed using the BNT77/BA27 and BNT77/BC05 antibody systems. BNT77, BA27, and BC05 were gifts of Takeda (Osaka).

In Western blotting the O443 antibody against the C-terminal 20 residues of APP was used for immunodetection of APP, CTF, and CTF.
