REAGENTS/MATERIAL: TPKI and TPK II were pruified from bovine brain as described in (Ishiguro et al, Takahashi et al.)
Human recombinant tau-441 purchased from Panvera.
Tau peptide K2 (191-224), partial phosphopeptides of tau pS202, pT205, pS208 (CPGDPGTPGSPRSRTPSL), pS210 (CPGDPGTPGSRSPRTPSL) and pS208/210 (CPGDPGTPGSPRSPRTPSL) were chemically synthesized employing solid-phase method and Biosearch Model 9500 or ABI Model 431A peptide synthesizer.
The antibodies pS199 and pT205 as described in (Ishiguro et al.)
The antibodies against pS208, pS210, pS208/210, and TTK-C were diluted to 15 ug/ml.
FUTURE DIRECTION: in vivo study on the precisely timed appearance of TTK in the regulation of the hyperphosphorylation of tau by TPKI/GSK-3b is needed. The physiological role of TTK needs further elucidation.
