REAGENTS/MATERIAL: All CSF samples were analyzed in duplicate using commercial ELISA kits for tau, for
phospho-tau (at threonine 181)
and for Aβ1–42 (all from Innogenetics, Ghent, Belgium).
We genotyped seven singlenucleotide polymorphisms (SNPs) extending alongthe tau gene and 87.2 upstream of exon 0. These SNPs were selected on the basis of previous studies showing that they cover major MAPT genetic variability, in order to compare their possible association with CSF tau markers. Fluorogenic allele-specific TaqMan® probes were usedfor genotypingrs1880753, rs1880756, rs1467967, rs242557, rs2471738 and rs7521, obtained from theTaqMan® SNP genotyping assays (Applied Biosystems).
