Findings

This variant was detected in one out of 376 control cases obtained from the KORA-Age cohort, based in Germany (Schulte et al., 2015). Limited information is available on this subject, but according to a description of the cohort as a whole, all individuals were Caucasian, and cognitively healthy at age 65 or older (Schulte et al., 2012).

Two heterozygous carriers of this variant were found in the gnomAD database (version 2.1.1, Oct 2021), one Latino/Admixed American and one European (non-Finnish).

Biological Effect

Mouse neuroblastoma cells expressing this variant secreted similar amounts of Aβ42 and Aβ40 compared with cells expressing wild-type APP, resulting in a similar Aβ42/Aβ40 ratio (Hsu et al., 2020). However, the PHRED-scaled CADD score, which integrates diverse information in silico, was above 20, suggesting a deleterious effect (CADD v.1.6, Oct 2021). Hsu and colleagues classified this variant as not pathogenic (Hsu et al., 2020).
