HWA 285
FDA Phase:
Discontinued
Role in Alzheimer's Disease:
General improvement in mild-moderately demented
Alzheimer's disease patients.
Pharmacological Role:
Adenosine uptake/phosphodiesterase inhibitor (? A
neuroprotective agent).
Side Effects:
Nausea (7% vs. 2% placebo); dizziness (6% vs. 3%); GI pain
(5% vs. 1%); headache (5% vs. 1%).
Evidence pro its efficacy:
A recent, pretty big, double-blinded placebo-controlled
multicenter study supported the use of propentofylline in
Alzheimer's and vascular dementia. The criteria that were
used included activities of daily living and cognitive
skills, both of which were quite statistically
significantly increased in the treated group.
Evidence con its efficacy:
A long term (72-week) PhaseIIIb clinical study yielded
disappointing results, with no treatment differences
between study population treated with propentofylline and
the population treated with placebo.
Dosage:
300 mg per day, three times daily.
Notes:
Development of propentofylline discontinued after
Phase IIIb clinical study. This record was last updated Dec
19, 2008.
